1、ICS 67.040 C 53 B 中华人民共和国国家标准食c 目口GB/T 5009.7-2003 代替GB/T5009.7-1985 中还原糖的测定Determination of reducing sugar in foods 2003-08-11发布2004-01-01实施中华人民共和国卫生变发布中国国家标准化管理委员GB/T 5009.7-2003 前言本标准代替GB/T5009. 7-1985V式中=X一试样中还原糖的含量(以某种还原糖计),单位为克每百克(g/100g) , A一碱性酒石酸铜溶液(甲、乙液各半相当于某种还原糖的质量,单位为毫克Cmg),m一试样质量,单位为克(g)
2、,V 测定时平均消耗试样溶液体积,单位为毫升(mL)。计算结果表示到小数点后一位。第二法离锺酸钢滴定法7 原理. ( 1 ) 试样经除去蛋白质后,其中还原糖把铜盐还原为氧化亚铜,加硫酸铁后,氧化亚铜被氧化为铜盐,以高健酸饵溶液滴定氧化作用后生成的亚铁盐,根据高锺酸何消耗量,计算氧化亚铜含量,再查表得还原糖量。50 GB/T 5009.7-2003 8 试剂8.1 碱性酒石酸铜甲液2称取34.639g硫酸铜(CuSO, 5H20),加适量水溶解,加0.5mL硫酸,再加水稀释至500mL,用精制石棉过滤。8.2 碱性酒石酸铜乙液2称取173g酒石酸御销与50g氢氧化销,加适量水溶解,并稀释至500
3、mL,用精制石棉过滤,贮存于橡胶塞玻璃瓶内。8.3 精fIjIJ石棉g取石棉先用盐酸(3mol/L)浸泡2d3 d,用水洗净,再加氢氧化纳溶液(400g/L)浸泡2 d3 d,倾去溶液,再用热碱性酒石酸铜乙液浸泡数小时,用水洗净。再以盐酸(3mol/L)浸泡数小时,以水洗至不呈酸性。然后加水振摇,使成细微的浆状软纤维,用水浸泡并贮存于玻璃瓶中,即可作填充古氏增塌用。8.4 高锺酸锦标准溶液(0.1000 mol/L)。8.5 氢氧化纳溶液(40g/L):称取4g氢氧化锅,加水溶解并稀释至100mL, 8.6硫酸铁溶液称取50g硫酸铁,加入200mL水溶解后,慢慢加入100mL硫酸,冷后加水稀释
4、至1000 mL, 8.7 盐酸(3mol/L):量取30mL盐酸,加水稀释至120mL。9 仪器9.1 25 mL古氏增塌或G4垂融瑞桶。9.2 真空泵或水泵。10 操作方法10.1 试样处理10. 1. 1 乳类、乳制品及含蛋白质的冷食类称取2.00 g 5. 00 g固体试样(吸取25. 00 mL50. 00 mL液体试样),置于250mL容量瓶中,加水50mL,摇匀后加10mL碱性酒石酸铜甲液及4mL氢氧化纳溶液(40g/L) ,加水至刻度,混匀。静置30min,用于燥滤纸过滤,弃去初滤液,滤液备用。10. 1. 2 酒精性饮料吸取100.0mL试样,置于蒸发皿中,用氢氧化纳溶液(4
5、0g/L)中和至中性,在水浴上蒸发至原体积的四分之一后,移人250mL容量瓶中。加50mL水,混匀。以下按10.1. 1自加10 mL碱性酒石酸铜甲液.起依法操作。10. 1. 3 含多量淀粉的食品z称取10.00 g 20. 00 g试样,置于250mL容量瓶中,加200mL水,在45(;水浴中加热1h,并时时振摇。冷后加水至刻度,混匀,静置。吸取200mL上清液于另一250mL 容量瓶中,以下按5.1. 1自加lOmL碱性酒石酸铜甲液.起依法操作。10. 1. 4 汽水等含有二氧化碳的饮料z吸取100.0mL试样置于蒸发皿中,在水浴上除去二氧化碳后,移入250mL容量瓶中,并用水洗涤蒸发皿
6、,洗液并人容量瓶中,再加水至刻度,混匀后,备用。10.2 测定吸取50.00mL处理后的试样溶液,于400mL烧杯内,加入25mL碱性酒石酸铜甲液及25mL乙液,于烧杯上盖一表面皿,加热,控制在4min内沸腾,再准确煮沸2min,趁热用铺好石棉的占氏瑞揭或G4垂融柑揭抽滤,并用60C热水洗涤烧杯及沉淀,至洗液不虽碱性为止。将古氏钳塌或垂融端纳放回原400mL烧杯中,加25mL硫酸铁溶液及25mL水,用玻棒搅拌使氧化亚铜完全溶解,以高锺酸御标准溶液c0/5 KMnO, O. 100 0 mol/L)J滴定至微红色为终点。同时吸取50mL水,加入与测定试样时相同量的碱性酒石酸翻甲液、乙液、硫酸铁溶
