7、biological activity on these specimens. 2 Apparatus 2.1 Corrosion-resistant soil container, approximately 50,8 cm in length and approximately 20,32 cm in width and with a depth of at least 12,7 cm. 2.2 Glass sheet or other suitable lid, to cover the soil container. 2.3 Sieve, with a 0,635 cm mesh sc

8、reen. 2.4 Means for maintaining the test soil at a constant temperature of 28 C 2 C, and with an adequate moisture content. 3 Test soil 3.1 Type A well-manured, fertile garden soil without a high clay content, rich in microbial life which decomposes cellulose, and capable of completely deteriorating

9、 the control specimen in 14 d. 3.2 Preparation of soil 3.2.1 Bring the moisture content of the soil to within the optimum limits by gradually adding water. NOTE The optimum moisture content of suitable soils is about 65 % to 70 % of their moisture retaining capacity, or 25 % to 30 % of their oven-dr

10、y weight. 3.2.2 Mix the water with the soil in such a manner as to avoid puddling. 3.2.3 Allow the soil to stand for 24 h then sift it through the sieve. 4 Control specimen A specimen which has in no way been proofed against microbial or fungal attack, but which is otherwise similar to the test spec

11、imens. SANS 5469:2009 Edition 2 4 5 Preparation of test specimens 5.1 Prepare test specimens of suitable dimensions to allow for subsequent tests and of shape that will allow uniform contact with the test soil. When relevant, subject the test specimens to the appropriate pre-treatment specified in t

12、he relevant standard for the required bacteria. 5.2 The quantity and dimensions of the test specimens shall be as specified in the relevant standard for the required bacteria. 6 Procedure 6.1 Submerge the specimens for 1 h in distilled water that contains 0,05 % of a non-ionic wetting agent. 6.2 Lay

13、 part of the test soil at the bottom of the container so as to form a 2,54 cm thick bed. 6.3 Place the test specimens and the control specimen flat on the soil bed, then cover them with a 2,54 cm thick layer of test soil. Cover the container (see 2.2). NOTE In cases where a breaking strength or othe

14、r test is to be used for subsequent evaluation, it is advisable not to cover areas which are later to be inserted in the jaws of a tensile strength testing machine or used for similar anchoring purposes. 6.4 Ensure that the correct moisture content and the correct temperature of the test soil are ma

15、intained throughout the period of incubation, i.e., unless otherwise specified, a period of 14 d. 6.5 On completion of the incubation period, remove the specimens and gently wash off all adhering soil particles. 6.6 Dry the specimens in air at room temperature and examine the control specimen to det

16、ermine whether the degree of microbiological activity was satisfactory. In cases where this is not evident, discard the specimens and repeat the test on a new set of test specimens with freshly prepared soil. 7 Evaluation 7.1 Visual examination Examine the test specimens for any apparent deteriorati

17、on and report the findings. 7.2 Other tests Determine the breaking strength or carry out other tests (or do both) in accordance with the method(s) specified in the relevant standard for the required bacteria.
