SANS 5811:2009 Edition 2 1 Pesticides Rearing and handling of the lesser grain borer (Rhizopertha dominica F.) 1 Scope This standard specifies a method for the rearing and handling of the lesser grain borer (Rhizopertha dominica F.). 2 Normative references The following referenced docum

9、ions prescribed in SANS 5808. 4 Rearing equipment 4.1 Rearing containers and lids, type A rearing containers and type A rearing container lids as described in SANS 5808, or any other suitable round, square or rectangular containers. 4.2 Sieves, two type A sieves as described SANS 5808 of nominal ape
10、rture sizes 850 m and 500 m, with the former placed on top of the latter, and that comply with the relevant requirements of SANS 3310-1. 5 Rearing medium Unground barley, heat-sterilized at 60 C 5 C for 30 min. NOTE Crushed maize or wheat may be used as an alternative. 6 Procedure 6.1 Place 10 mL of
11、 adult lesser grain borers on 1 kg of the rearing medium (see clause 5) in a rearing container (see 4.1). SANS 5811:2009 Edition 2 2 6.2 After 2 weeks, transfer the entire contents of the rearing container on to the sieves (see 4.2) and sift gently. The barley will be retained on the 850 m sieve, an
12、d the adult insects on the 500 m sieve. Retain the adults for use as laid down in 6.4. 6.3 Return the barley (containing the eggs) to the rearing container. The adults will emerge after approximately four weeks. 6.4 Rear new colonies by repeating the procedure in 6.1 to 6.3 (inclusive), but using fo
13、r each colony 5 mL of the adult grain borers retained in 6.2 and 5 mL of freshly-emerged adults (see 6.3).
