The responsible German body involved in its preparation was the Normenausschuss Wa

This standard has been published to implement the Water Framework Directive (WFD), Directive 2000/60/EC of the European Parliament and of the Council of 23 Octo

7、ber 2000 establishing a framework for Community action in the field of water policy1). ISO 11348 consists of the following parts, under the general title Water quality Determination of the inhibitory effect of water samples on the light emission of Vibrio fischeri (Luminescent bacteria test): Part 1

19、 standard differs from DIN EN ISO 11348-3:1999-04 as follows: a) The scope has been extended to include: sea water; sediment eluates (fresh water, brackish and sea water); single substances, diluted in water. b) Clause 4 “Interferences” has been revised: the oxygen concentration of readily biodegrad

20、able nutrients is no longer specified; reference is now made to Bibliography 1; specifications regarding interferences due to samples with extreme pH values have been included; when testing salt-water samples, possible inferences as a result of stimulation effects of bioluminescence are to be taken

21、into account. c) Clause 5 “Reagents and materials”: the concentration of the 3,5-Dichlorophenol reference substance has been changed and the purity has been specified; commercially available reference substance solutions may now be used. d) Modifications relating to the preparation of the procedure:

22、 apparatus: the permissible temperature deviation within one test has been changed from 0,2 C to 0,3 C; sampling: samples may now be stored at 18 C for periods of up to two months (instead of two weeks); sample preparation: the oxygen concentration is to be measured in accordance with ISO 5814 and s

23、amples having an oxygen concentration of less than 3 mg/l are to be oxygenated by aeration or stirring; e) Modifications relating to the procedure: two parallel determinations have been specified for the tests; the permissible temperature of the stock suspension has been changed from 3 C 3 C to 4 C

24、3 C. f) A further method for the calculation of the concentration-effect relationship has been included (e.g. as in ISO/TS 20281). g) In Annex B, the volumes of the test and control batches have been modified and a dilution scheme has been included. h) Annex D “Testing salt water samples with the lu

37、n for Standardization (ISO) and has been taken over as EN ISO 11348-3:2008 by Technical Committee CEN/TC 230 “Water analysis” the secretariat of which is held by DIN. This European Standard shall be given the status of a national standard, either by publication of an identical text or by endorsement

41、as a EN ISO 11348-3:2008 without any modification. Introduction The measurements specified in ISO 11348 can be carried out using freshly prepared bacteria, as well as freeze-dried or liquid-dried bacterial preparations. Standardized work carried out by DIN Normenausschuss Wasserwesen and ISO/TC 147/

42、SC 5/WG 1 has shown that, in special cases, these different techniques may give different results, especially in the presence of heavy metals. Such varying sensitivity is caused by differences in media composition used in the preparation of freeze-dried or liquid-dried bacteria. These protective med

43、ia influence the bioavailability of toxicants and/or the light emission of luminescent bacteria. This means that the origin and type of preparation need to be taken into account when interpreting the results. This may be difficult sometimes, as freeze-dried and liquid-dried bacteria may be obtained

44、from different suppliers. This, in turn, can mean that the composition is not known in detail and therefore cannot be interpreted by the user. For this reason, in addition to toxicity measurements with liquid-dried bacteria (ISO 11348-2) and freshly prepared bacteria (ISO 11348-1), a procedure with

45、freeze-dried bacteria is described in this part of ISO 11348, the performance of which can be interpreted by the user in every detail. The laboratories responsible for the results have the opportunity to select the most suitable technique based on expert judgement and information about the water sam

46、ple to be tested. DIN EN ISO 11348-3:2009-05 EN ISO 11348-3:2008 (E) 4WARNING Persons using this part of ISO 11348 should be familiar with normal laboratory practice. This standard does not purport to address all of the safety problems, if any, associated with its use. It is the responsibility of th

48、e methods for determining the inhibition of the luminescence emitted by the marine bacterium Vibrio fischeri (NRRL B-11177). This part of ISO 11348 specifies a method using freeze-dried bacteria. This method is applicable to: waste water; aqueous extracts and leachates; fresh water (surface and grou
