SANS 5302:2009 Edition 3 1 Pesticides Rearing and handling of the skin and hide beetle (Dermestes maculatus De Geer) 1 Scope This standard specifies a method for the rearing and handling of the skin and hide beetle (Dermestes maculatus De Geer). 2 Normative references The followi

9、 the rearing conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Brush, soft hairbrush. 4.2 Rearing containers and lids, type A rearing containers and type A rearing container lids as described in SANS 5808, or any other suitable round, square or rectangular containers. 4.3 Sand, sieved (see
10、 4.4) and sterilized by heating at 70 C for at least 1 h or until dry. 4.4 Sieve, type A sieve as described in SANS 5808 of nominal aperture size 1,7 mm, and that complies with the relevant requirements of SANS 3310-1. 5 Media 5.1 Prepare oviposition and rearing medium as follows: a) Cut unbruised u
11、nsalted beef that is free from sinew and fat into strips approximately 50 mm wide and 40 mm thick and of convenient length. Dry the strips before a rapidly revolving fan until the meat is dry and hard on the outside but still moist on the inside. SANS 5302:2009 Edition 3 2 b) Divide a tin of canned
12、corned beef into four pieces and use two of the pieces at a time. 5.2 Saturate pieces of sterile cotton wool with tap water and place in disposable plastics containers of approximate height 60 mm and diameter 110 mm. 6 Procedure 6.1 Place approximately 250 g of the oviposition and rearing medium (se
13、e 5.1) and a water container (see 5.2) on a 20 mm thick layer of sand (see 4.3) in a rearing container (see 4.2). 6.2 Place in the rearing container approximately 600 adult skin and hide beetles. NOTE The females will deposit their eggs on the medium and the sand. 6.3 Remove the medium after one wee
14、k, shaking off the beetles by sharply tapping the meat against the side of the rearing container. 6.4 Sift the contents of the rearing container through the sieve (see 4.4), and return the sand to the container. The adults and coarse debris will be retained on the sieve. 6.5 Separate the adults from
15、 the debris using the brush (see 4.1) and retain them for use in 6.8. 6.6 Place the medium containing the eggs (see 6.3) and a fresh water container on the sand in the rearing container (see 6.4). 6.7 Examine the rearing container regularly and replenish with medium and fresh saturated cotton wool a
16、s and when required. The adults will emerge after approximately eight weeks. 6.8 Rear new colonies by repeating the procedure in 6.1 to 6.7 (inclusive), but using for each colony 300 of the adults retained in 6.5 and 300 of the freshly-emerged adults (see 6.7). 6.9 Destroy the excess adults from 6.5
