SANS 5458:2008 Edition 4 1 Pesticides Rearing and handling of the German cockroach (Blatella germanica (L.) 1 Scope This standard specifies a method for the rearing and handling of the German cockroach (Blatella germanica (L.). 2 Normative references The following referenced documents ar

SANS 5808, Pesticides General conditions and equipment for the rearing and handling of test insects. 3 Rearing conditions Maintain the rearing conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Feeding dishes, type B plastics dishes as described in SANS 5808.
9、 4.2 Rearing containers and lids, type B rearing containers and type A rearing container lids as described in SANS 5808. 4.3 Shelters, cockroach shelters as described in SANS 5808. 4.4 Sieve, type A sieve as described in SANS 5808 of nominal aperture size 4,0 mm. 5 Rearing media 5.1 Finely ground do
10、g biscuits contained in a feeding dish (see 4.1). 5.2 Cotton wool saturated with water that contains methyl-4-hydroxy benzoate (0,9 g/L), and placed in a feeding dish (see 4.1). SANS 5458:2008 Edition 4 2 6 Procedure 6.1 Place about 2 000 (20 mL of) freshly-emerged first nymphal instars of the Germa
11、n cockroach (see 6.6) in a clean rearing container (see 4.2) and supply with the rearing media (see 5.1 and 5.2) and the shelters (see 4.3) arranged in two tiers (not more than 20 shelters per rearing container). 6.2 After 12 weeks, anaesthetize the insects in the rearing container using carbon diox
12、ide, and separate the females with othecae, the females without othecae, the males, and the nymphs manually. Destroy any nymphs found at this stage. 6.3 Return females without othecae to the rearing container and examine them after one week for the development of othecae. Destroy females still witho
13、ut othecae, and transfer the others to the rearing container described in 6.5. 6.4 Place males in a separate rearing container and supply with the rearing media and the shelters. NOTE Only males are used for the evaluation of pesticides. 6.5 Place females with othecae (see 6.2) in a clean rearing co
14、ntainer and supply with the rearing media and the shelters. 6.6 After one week, and each consecutive week thereafter, anaesthetize the adult breeding stock (see 6.5) in the rearing container using carbon dioxide, and use the sieve (see 4.4) to separate the adults from the first nymphal instars. Repe
15、at this procedure until all the adults have died. 6.7 After the nymphs and adults are separated (see 6.6), discard any dead adults and return the others to the rearing container. Supply with fresh rearing media. 6.8 Supply the colony with fresh rearing medium once every week. NOTE If the colony should become too large to be contained in one rearing container, the insects may be distributed over two or more containers. 6.9 Rear new colonies by repeating the procedure in 6.1 to 6.8 (inclusive) for each colony.
