SANS 5808, Pesticides General conditions and equipment for the rearing and handling of test insects. 3 Rearing conditions Maintain the rearing conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Rearing containers and lids, type A rearing
9、containers and type A rearing container lids as described in SANS 5808. 4.2 Sieve, type A sieve as described in SANS 5808 of nominal aperture size 850 m. 5 Rearing medium A mixture of the following: a) 1 125 g cake flour, heat-sterilized at a temperature of 60 C 5 C for 30 min; b) 1 125 g coarse whe
10、at flour, obtained by sifting ground wheat through the sieve (see 4.2) and heat-sterilizing the fine fraction at a temperature of 60 C 5 C for 30 min; c) 270 g full-cream milk powder; and d) 180 g fishmeal, sifted (see 4.2) and heat-sterilized at a temperature of 60 C 5 C for 30 min. 11、Edition 4 2 6 Procedure 6.1 Place 100 mL of adult flour beetles (about 16 000 beetles) on top of the rearing medium (see clause 5) contained in a rearing container (see 4.1). 6.2 After 14 d separate the rearing medium and eggs from the adults by sieving the entire contents of the rearing container t
12、hrough the sieve (see 4.2). Retain the adults that remain behind on the sieve for use in 6.5. 6.3 Transfer the rearing medium and eggs to a fresh rearing container. 6.4 The adult stage is reached after approximately 8 weeks to 10 weeks and adults are separated by sieving as in 6.2. 6.5 Start a new colony by repeating the procedure in 6.1 to 6.4 (inclusive), but using 50 mL of the flour beetles retained in 6.2 and 50 mL of the flour beetles obtained in 6.4.
