SANS 5808, Pesticides General conditions and equipment for the rearing and handling of test insects. 3 Rearing conditions Maintain the rearing conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Cardboard sheet, thin. 4.2 Filter paper circles, medium gra
9、de, and of diameter 150 mm. 4.3 Rearing containers and lids, type B rearing containers and type A rearing container lids as described in SANS 5808. 4.4 Sieve, two type A sieves as described in SANS 5808 of nominal aperture size 840 m. 5 Rearing media 5.1 Oviposition medium. Heat sterilize 500 g each
10、 of white and yellow mealie meal at 60 C 5 C for 30 min and allow to cool. Mix the white and yellow mealie meal and add 50 g of brewers yeast, and 20 g of full-cream milk powder. Mix thoroughly in the rearing container (see 4.3). 5.2 Sugar solution, 5 g/100 mL of sugar in water. SANS 5809:2008 Editi
11、on 2 2 5.3 Wheaten bran, 500 g of sifted wheaten bran which has been heat sterilized at 60 C 5 C for 30 min and allowed to cool. 6 Procedure 6.1 Saturate two circles of the filter paper with sugar solution (see 5.2) and place side-by-side on the oviposition medium (see 5.1) in the rearing container
12、(see 4.3). 6.2 Place 7,5 mL of adult tobacco beetles on each of the filter paper circles (see 4.2) (a total of approximately 3 250 beetles), and cover each circle with a V-shaped roof made from the cardboard sheet (see 4.1). Cover the container with the lid, and place in the insect rearing room (see
13、 clause 3). 6.3 After 8 d, remove the filter paper and cardboard and sieve the contents of the rearing container to separate the eggs and the medium from the adult beetles. Destroy the adult beetles that remain on the sieve. 6.4 Add the wheaten bran (see 5.3) to the sieved medium containing the eggs
14、 and mix lightly (until the appearance is mottled) to form a rearing medium (containing the eggs). Return it to the rearing container. Replace the lid of the rearing container and keep the container in the insect rearing room. 6.5 After seven weeks, use a blunt object to break the crust that has for
15、med on the rearing medium into pieces of size approximately 50 mm 50 mm. 6.6 Place two fresh circles of filter paper saturated with sugar solution on the rearing medium (see 6.4). Cover each circle with a V-shaped roof made from the cardboard sheet. Replace the lid of the rearing container and keep the container in the insect rearing room. 6.7 The first adults should emerge after approximately eight weeks and will collect on the sides of the container and on the V-shaped pieces of cardboard. 6.8 Remove the adults and start a new colony, following the procedure in 6.1 to 6.7 (inclusive).
