28、lulosic fibres to deterioration by the action of a cellulolytic fungus. The method is of particular importance in testing fabrics designed for outdoor use. The test is of moderate severity and is not recommended for treated fabrics intended for use under conditions involving contact with soil. (See
29、CAN/CGSB-4.2 No. 28.3-M.) The testing and evaluation of a product against this method may require the use of materials and/or equipment that could be hazardous. This document does not purport to address all the safety aspects associated with its use. Anyone using this method has the responsibility t
30、o consult the appropriate authorities and to establish appropriate health and safety practices in conjunction with any existing applicable regulatory requirements prior to its use. PRINCIPLE The fabric is leached with water to remove any fungicidal substance likely to be rapidly removed by wetting,
31、and to ensure that all specimens be in uniform condition with respect to shrinkage. Specimens taken from the leached fabric are subjected to the action of a cellulolytic fungus under controlled conditions, and in the absence of any source of assimilable carbon other than that present in the test mat
32、erial. The resistance of the fabric is judged by the loss in strength resulting from exposure to the organism for a period of time known to cause severe loss of strength in cellulosic materials that have not received any protective treatment. 37、sm: the fungus is grown on agar slants in test tubes, and spore suspensions are made in sterile distilled water as required for this test. Subcultures should be made on filter paper placed on mineral salts agar at sufficiently frequent intervals to maintain viable pure cultures. Cultures, under refr
38、igeration, may be kept as long as six to nine months. Salts - Agar culture medium: a typical satisfactory formula is: Sodium nitrate (NaN03) 2.0 g Magnesium sulphate (MgS04) 0.5 g Potassium chloride (KCl) 0.5 g Ferric sulphate (Fez( SO&- H20) Potassium orthophosphate monohydrogen (K2“04) Yeast extra
39、ct 0.02 g 0.01 g 1.2 g Potassium orthophosphate dihydrogen (KH2P04) 0.14 g Agar 15.0 g Distilled Water 1.0 L Adjust pH to 7.2 f 0.1 with hydrochloric acid, plus sodium hydroxide if necessary, and store in flask. Autoclave: an autoclave capable of being operated at a steam pressure of 103 to 117 kPa
40、(gauge) and an exhaust temperature of 121 to 123OC. these being the sterilizing conditions used throughout. Incubator: an incubator room or cabinet maintained at 30 f 2C. Petri dishes: 150 mm in diameter. Leaching Apparatus This shall consist of a reservoir of water at 24 f 3OC from which water is p
41、assed to a suitable vessel containing the fabric to be leached. The size of this vessel will depend on the size of the sample to be leached. The rate of flow of water through the vessel should be such as to provide approximately four changes of water per hour. The water used must not contain any mat
42、erial that may react with any of the constituents of the sample being tested (Note 2). Wherever possible, distilled or demineralized water shall be used. The pH of the water shall not be less than 5.0 nor greater than 8.0. Low power binocular microscope (SOX). TEST SPECIMENS Fabrics - A portion appr
43、oximately 200 x 400 mm, with the shorter dimension parallel to the warp, shall be cut from the test sample and leached for 24 h (Note 3). Nine warpwise strips, each approximately 200 x 35 mm, shall be cut from the leached portion, ravelled to a width of exactly 25 mm, and cut into two warpwise strip
44、s each 100 mm long. The nine 100 mm long specimens from the upper half of the 200 x 400 mm piece shall be kept together in one lot and the nine specimens from the lower half in Note 2: An example of the use of an unsuitable type of water is to employ a water containing alkaline impurities to leach a
45、 sample carrying a phenolic type fungicide. Depending on the degree of alkulinity, solubilization of the fungicide y the alkali in the water will occur through the formation of a soluble phenate, and the loss thus occasioned would not represent a loss through water (pH 6-8) solubility of the origina
46、l phenolic compound. Note 3: To facilitate wetting by the spore suspension, it is desirable that the inoculation of the specimens (section 6) be proceeded with immediately following leaching, before the specimens have become dry. Where drying has occurred, the specimens should be dampened by being t
47、horoughly sprayed with distilled water and allowed to stand for 1 to 2 h,flat, on a non- absorbent sulface in a closed container, to avoid dessication. Yarns - Forty-two 500 mm lengths shall be taken from the sample and wound into loops approximately 50 mm long, and leached for 24 h (Note 3). Twenty-one loops are used as control specimens, the other twenty-one being subjected to the action of the organism.
