SANS 5299:2008 Edition 3 1 Pesticides Rearing and handling of the Mediterranean flour moth (Ephestia kuehniella Zeller) 1 Scope This standard specifies a method for the rearing and handling of the Mediterranean flour moth (Ephestia kuehniella Zeller). 2 Normative references Th

SANS 5808, Pesticides General conditions and equipment for the rearing and handling of test insects. 3 Rearing conditions Maintain the rearing conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Aspirator, as described in SAN
9、S 5808. 4.2 Brush, soft hairbrush. 4.3 Filter paper, of diameter 140 mm. 4.4 Graduated 5 mL centrifuge tube. 4.5 Oviposition containers, disposable plastics containers as described in SANS 5808. 4.6 Rearing containers and lids, type B rearing containers and type A rearing container lids as described
10、 in SANS 5808. 4.7 Sieves, two type A sieves as described in SANS 5808 of nominal aperture sizes 425 m and 250 m, with the former placed on top of the latter. SANS 5299:2008 Edition 3 2 5 Rearing medium Sterilize 1 kg unsifted wheaten meal by heating it at a temperature of 70 C for 30 min, allow it
11、to cool, and thoroughly mix it with 50 g dried live brewers yeast. 6 Procedure 6.1 Use the aspirator (see 4.1) connected to a suction apparatus operated at a very low airflow to collect 600 freshly-emerged Mediterranean flour moths (see 6.7), anaesthetize them with carbon dioxide, and transfer them
12、to an oviposition container (see 4.5). 6.2 Place the oviposition container upside-down on a piece of filter paper (see 4.3) on the bottom of a rearing container (see 4.6). 6.3 Remove the filter paper with the adhering eggs after 4 d. Transfer the eggs from the filter paper on to the sieves (see 4.7)
13、 using the brush (see 4.2). 6.4 Sift the eggs. The moth scales will be retained on the 425 m sieve, and the eggs on the 250 m sieve. 6.5 Destroy the moths which were used for oviposition. 6.6 Measure 0,5 mL of the eggs in the graduated centrifuge tube (see 4.4), distribute them on the rearing medium
14、 (see clause 5) in a rearing container, and cover the container with the lid. Repeat with further 0,5 mL volumes of the eggs and further containers. 6.7 The adults will emerge after six weeks to seven weeks. 6.8 Rear new colonies by repeating the procedure in 6.1 to 6.7 (inclusive) for each colony.
