SANS 5808, Pesticides General conditions and equipment for the rearing and handling of test insects. 3 Rearing conditions Maintain the conditions prescribed in SANS 5808. 4 Rearing equipment 4.1 Rearing containers and lids, type A rearing containers and type A rearing
9、 container lids as described in SANS 5808. 4.2 Sieves, two type A sieves as described in SANS 5808 of nominal aperture sizes 1,70 mm and 850 m, with the former placed on top of the latter. 5 Rearing medium Approximately 4 500 g wheat, heat-sterilized at a temperature of 60 C 5 C for 30 min. 6 Proced
10、ure 6.1 Place 160 mL of adult granary weevils (about 24 000 weevils) on top of the rearing medium (see clause 5) contained in a rearing container (see 4.1). 6.2 After 14 d sieve the entire contents of the rearing container gently through the combination of sieves (see 4.2) to separate the adults (re
11、tained on the top sieve), the debris (retained on the bottom sieve), and the infested wheat. Discard the debris, and retain the adults for use in 6.4. SANS 5460:2008 Edition 4 2 6.3 Return the infested wheat (which has passed through both sieves) to the rearing container. Weevils will emerge after six weeks to eight weeks. 6.4 Start a new colony by repeating the procedure in 6.1 to 6.3 (inclusive), but using 80 mL of the granary weevils retained in 6.2 and 80 mL of the weevils obtained in 6.3.
