This specification covers to detect anticholinesterase agents such one type of detector ticket designed as GB and VX. 2. REQUIREMENTS 3.1 Materials and components. 3.1.1 Materials. Al1 materials cited on Drawing 5-77-2510 and subsidiary drawings shall-conform

All components of the detecto

13、r ticket packet assembly shall conform to Drawing 5-77-2510 and subsidiary drawings . 3.2 Manufacture and assembly. The detector ticket packet shall be manu- factured and assembled in accordance with Drawing 5-77-2510 and subsidiary drawings . 3.3 Perforaance requirements for ambient stored detector

14、 tickets. 3.3.1 Cholinesterase recovery before aging. No less than 0.20 and no more than 0.80 cholinesterase units Ber disc shall be recovered before aging when tested as specified in 4.4.4.1, 3.3.2 VX sensitivity before aging. Detector tickets shall have no color change before aging when tested as

15、specified in 4.4.4.2, 3.3.3 Blank before aging. After 3 minutes, upon challenging with pure water hue 7.5BG, lOBG, 2.5B, 5B, before aging, the enzyme disc (clipped end) shall develop one of the following Munsell colors when tested as specified in 4.4.4.3: 7.5B, 103; value 7 to 9; chroma 2 or darker.

16、 3.4 Performance requirements for accelerated aging conditions . The detector ticket shall meet the following requirements after accelerated aging for 9 con- tinuous days at 90 - + 2OC. 3.4.1 Cholinesterase recovery after aging. After accelerated aging, no less than 0.20 and no more than 0.80 cholin

17、esterase units per disc shall be recov- - ered when tested as specified in 4.4.4.4. 3.4.2 VX sensitivity after aging. Detector tickets shall have no color change after accelerated aging when tested as specified in 4.4.4.5. 3.4.3 Blank after aging. After 3 minutes, upon challenging with pure water af

18、ter accelerated aging, the enzyme disc (clipped end) shall develop one of the following Munsell colors when tested as specified in 4.4.4.6: hue 7.5BG, lOBG, 2.5B or 5B; value 7 to 9; chroma 2 or darker. 3.5 pH of water extract. Detector tickets shall have a pH between 8.5 and that of the water befor

19、e extraction when tested as specified in 4.4.4.7. 3.6 Tear opening. Detector tickets shall be removable from the foil pack without distorting the ticket or using a tool or finger to enlarge the opening when tested as specified in 4.4.4.8. 3.7 Leakage. There shall be no visible bubble leakage any pla

20、ce on the tickets foil pack whentested as specified in 48484.90 3.8 Overlay heat seal. The overlay shall strip off so that no more than 1/16 inch of overlay material extends inside the inner edge of the heat seal when tested as specified in b.4.4.10. 3 Provided by IHSNot for ResaleNo reproduction or

23、 Responsibility for inspection. Unless otherwise specified in the con- tract or purchase order, the contractor is responsible for the performance of all inspection requirements as specified herein. in the contract or purchase order, the contractor ;a4y use his own or any other facilities suitable fo

The inspection requirenents specified herein are classified as follows: (a) Preproduction inspection (see 4.3) (b) Quality conformance inspection (see 4.4) 4.3 Preproduction inspection. 4.3.1 Lotting. A preproduction

26、lot of no less than 525 detector tickets shall be manufactured using the same methods, materials, equipment, and pro- cesses of manufacture as will be used during regular production (see 6.2). 4 3 .2 Sampling and inspection procedure . 4.3.2.1 For examination. Sampling and inspection procedure for e

27、xamination shall be as specified in 4.4.2.1 and 4.4.3.1. 4.3.2.2 For test. Sampling shall be conducted in accordance with table I. Tests shall be conducted as specified in 4.4.4. 4 . 4 Quality conformance inspect ion . 4.4.1 Lotting. A lot of tickets shall consist of the detector tickets impregnated

37、3.2 For test. 4.4.4 Tests (see 6.7). Inspection procedure shall be as specified in table I. Water in accordance with ASm Di193 and reagent grade chemicals shall be used throughout the tests. Where applicable, blank dete.minations shall be run and corrections applied where significant. Tests shall be

38、 conducted as follows: 4.4.4.i Cholinesterase recovery before aging. (a) PH stat solution. To prepare 1 liter of pH stat solution, dissolve 8.120 grams (g) of magnesium chloride (O.O4M), 5.850 g of sodium chloride (O.lOM), and 0.730 Q of acetylcholine chloryde or 0.909 g of acetyl- choline bromide i

39、n 900 milliliters (mL) of deionized water having a pH between 5.0 and 6.0 (the pH of final solution will be approximately 4.5). Adjust the final volume to 1 liter with deionized water. (Solution is stable for 10 con- secutive calendar days if kept refrigerated at 5“ - + 5C.) 6 Provided by IHSNot for

Equilibrate this solution in a water bath to 37“ + 1C. To a 20-mL beaker ad

41、d 2.0 mL of deionized water having a pH between 7.0 and 7.2. Peel off one eel acetylcholinesterase enzyme-impregnated paper disc from the clipped end of the ticket and add to the beaker. (Scrape off as much of disc as possible from around seal area and also add these scrapings to the beaker.) Break

42、up the paper disc with a glass rod and then stir vigorously at room temperature for 5 minutes with a magnetic stirrer. While this is mixing, adjust the pH of the pH stat solution to 8.0 using an automatic, titrator (pH stat type), using 0.004N NaOH and maintaining 37“ + 1C. After tenperature and pH

Dilute a quantity of VX of known purity, based on an assay performed within the last 60 days (see 6.6 for a suggested method), to 100 mL in a volumetric flask with isopropyl alcoho

47、l so that the solution contains 10 microliters of pure VX (Stock Solution A). Dilute 0.1 mL of Stock Solution A to 100 mL in a volumetric flask with deionized water having a pH of between 5.0 and 6.0 (Stock Solution B). 2 mL of Stock Solution B to 10 mL with deionized water in a volumetric flask to
